EB病毒及其多种因素的相互关系在鼻咽癌发生发展中的作用

Epstein Barr virus and the interaction of the multiple related factors for the occurrence and progression of nasopharyngeal carcinoma

目的:观察EB病毒膜蛋白(LMP-1)、信号传导与转录激活因子signal(transductionandactivatorsoftranscription,STATs)Stat-1及Stat-3在鼻咽良、恶性病变中的表达,探讨其间相互关系及其在鼻咽癌发生发展中的作用。方法:选择1999-01/2001-12广东医学院第二附属医院耳鼻喉科和农垦医院耳鼻喉科人鼻咽黏膜慢性炎29个和鼻咽癌组织60个。采用免疫组化技术检测29个慢性鼻咽炎,60个鼻咽癌组织中LMP-1,Stat-1,Stat-3蛋白的表达。结果:29例慢性鼻咽炎LMP-1,Stat-1,Stat-3阳性率分别为17%(5/29),10%(3/29),41%(12/29);60例鼻咽癌组织中LMP-1,Stat-1,Stat-3蛋白的阳性表达率分别为40%24/60),28%17/60),86%52/60),LMP-1(((与Stat-3蛋白表达呈明显正相关(r=0.7382,P<0.01),而LMP-1与Stat-1蛋白表达无相关性(r=0.2186,P>0.05)。结论:Stat-1,Stat-3在鼻咽癌中有异常表达。鼻咽癌组织中EB病毒感染可能与Stat-3的活化有关。

AIM:To investigate the expression of Epstein Barr(EB) virus latent membrane p rotein 1(LMP 1),signal transduction and activators of transcription(STATs,Stat 1 and Stat 3) in the nasopharyngeal benign and malignant lesions,and probe in to the correlation of LMP 1 with Stat 1,Stat 3 and their role in the occurren ce and development of nasopharyngeal carcinoma(NPC). METHODS:From January 1999 to December 2001,29 cases of human chronic nasophary ngitis with mucous membrane and 60 cases of NPC were selected from Department of Otorhinolaryngology in the Second Affiliated Hospital of Guangdong Medical Coll ege and Department of Otorhinolaryngology,Nongken Hospital.Immunohistochemical m ethod was used to examine the expression of LMP 1,Stat 1 and Stat 3 in all th e nasopharyngeal tissues. RESULTS:The positive rates of LMP 1,Stat 1 and Stat 3 were 17%(5/29),10%( 3/29) and 41%(12/29) respectively in the 29 cases of chronic nasopharyngitis,an d 40%(24/60),28%(17/60) and 86%(52/60) respectively in the 60 cases of NPC.Th e expression of LMP 1 was positively related to the expression of Stat 3 in NP C tissues(r=0.738 2,P< 0.01),but there was no significant correlation between th e expression of LMP 1 and Stat 1(r=0.218 6,P >0.05). CONCLUSION:The expressions of Stat 1 and Stat 3 are abnormal in NPC tissues. EB virus infection may be involved in the activation of Stat 1 and Stat 3 in N PC tissues.
