复方硒多糖胶囊对内皮细胞凋亡的影响 被引量:3

摘要 目的 用 2 ,2′ -盐酸脒基丙烷 [2 ,2′ -azobis- (2 -amidinopropane)hydrochloride ,AAPH]诱导血管内皮细胞凋亡 ,观察复方硒多糖胶囊是否具有抗凋亡的作用。方法 MTT测定并分析细胞损伤状况 ,倒置相差显微镜观察细胞形态 ,DNA提取及琼脂糖凝胶电泳观察凋亡特征性梯带 ,流式细胞仪检测细胞凋亡。结果 经 10 μmol·L-1复方硒多糖溶液预处理后 ,由AAPH诱导的内皮细胞凋亡受到明显抑制。结论 复方硒多糖胶囊可抑制氧化所致的内皮细胞凋亡 ,具有抗动脉粥样硬化的作用。 OBJECTIVE To investigate whether Compound selenium and polysaccharide capsules can prevent the apoptosis of endothelial cells caused by 2,2′-azobis-(2-amidinopropane)hydrochloride (AAPH). METHODS The damnificational level and the morphology of cells were measured by MTT assay and inverted microscopy, respectively. The apoptosis of cells was detected by DNA agarose gel electrophoresis and flow cytometer. RESULTS After pretreatment with the solution of Compound selenium and polysaccharide at the concentration of 10 μmol·L -1, the apoptosis of endothelial cells caused by AAPH was obviously inhibited. CONCLUSION Compound selenium and polysaccharide capsules can prevent the apoptosis of endothelial cells.
