摘要
目的通过托吡酯及巴氯芬干预后致痫大鼠海马神经元特异性烯醇化酶(NSE)蛋白表达的变化,进一步探明托吡酯及巴氯芬在致痫脑损伤及神经保护方面所起的作用。方法在立体定位仪下,将海人藻酸(kainicacid,KA)注射至大鼠杏仁核制备癫痫模型。将大鼠随机分为正常组、致痫组及药物干预组,分别于不同时间取材进行脑组织NSE蛋白检测。结果与对照组相比KA致痫后脑组织的NSE表达明显下降,与KA组相比托吡酯及巴氯芬干预后NSE蛋白表达增加(P<0.05)。结论KA致痫后NSE蛋白表达下调,托吡酯及巴氯芬干预后NSE蛋白表达上调。
Objective By comparing the changes of NSE protein expression in hippocampus of epilepsy rats induced by kainic acid (KA) after intervened by topirate and baclofen to explore the effect of topirate and baclofen on brain injury and neural protection in induced epilepsy rats. Methods Under three-dimensional orientator, kainic acid was injected into amygdaloid nucleus of rats to prepare epilepsy rats model. These rats were randomly divided into normal group, KA group, topiramate group and baclofen group, the expression of NSE was determined by immunohistochemistry methods. Results Comparing to that in control group, the NSE expression in KA group significantly decreased. The NSE expression in topiramate and baclofen group increased comparing to that in KA group (P<0.05).Conclusions NSE protein expression in epilepsy rats decreased induced by KA and upregulated induced by topirate and baclofen.
