小鼠HSV-2阴道炎粘膜上皮细胞凋亡和Fas/FasL异常表达关系的研究

摘要 目的 :探讨 HSV- 2在体内诱导细胞凋亡的效果、形态学特点和 Fas蛋白的表达。方法 :用 HSV- 2 333株感染小鼠阴道 ,光镜下进行原位观察 ,TUNEL末端标记染色显示凋亡的细胞 ,免疫组化技术检测 Fas蛋白的表达。结果 :感染后的第 1天粘膜上皮内即出现大量的凋亡细胞 ,第 3~ 6天凋亡细胞的数量及在上皮内的分布范围达最高水平。凋亡细胞被 TUNEL标记染成棕褐色 ,经免疫组化技术证实凋亡细胞有 Fas蛋白的表达。结论 :HSV- 2 333株阴道感染可同时诱导细胞坏死及凋亡 ,Fas蛋白的表达参与了细胞凋亡的发生。 Objective:To explore the effect of HSV-2 strain 333 on induction of apoptosis and its morphological characters and the expression of Fas protein in vaginal epithelia in mice infected with the virus intravaginally. Methods:Mice were killed day 1-12 days after infection(P. I.).The vaginas were taken and fixed in 10% neutral formalin for paraffinembeding section and TUNEL staining and immunohistochemitry technique to demonstrate apoptosis . Results: Numerous apoptotic cells appeared in the epithelia on the first day of infection,the number of the apoptotic cells reached to a high level on day 3-6 P.I.. Apoptosis cells are stained by TUNEL staining and had the expression of Fas protein.At the same time,the necrotic areas and herpetic vesicles in the epithelia could be found to be restricted only in some areas and were surrounded by the apoptotic areas. Conclusion:The results showed that the HSV-2 strain 333 could induce apoptosis and necrosis of vaginal mucosal epithelia at the same time in mice.There is the expression of Fas protein in apoptosis cell. The apoptosis might play an important role in limiting both the range of viral spreading and the production of progeny virus.
