摘要
目的 :探讨肿瘤坏死因子 ( TNFα)对人鼻咽癌 CNE3细胞生长相关基因表达的影响。方法 ;体外培育的人鼻咽癌低分化上皮细胞株 CNE3,通过 MTT细胞毒试验确定TNFα的最佳抑制剂量 ,用免疫组织化学技术观察 TNFα作用 48h后 ,CNE3细胞中增殖相关的几种蛋白表达的变化。结果 :TNFα可下调 PCNA、Ki- 67、p2 1 ras、mtp5 3等蛋白的表达 ,增强 p2 1 WAF1蛋白表达。结论
Objective:To study the influence of tumor necrosis factor alpha (TNFα) on proliferation related gene expression of human nasopharyngeal carcinoma cell line CNE3 in vitro. Methods: CNE3 cell line of human nasopharyngeal carcinoma were cultured by cell culture technique. Immunocytochemistry was used to determine the expressions of PCNA,Ki-67,p21ras,mtp53 genes of CNE3 cells altered by TNFα. Results:There are expressions of PCNA,Ki-67,mtp53 gene proteins in most nucleolus of CNE3 cells, with the positive percentage of PCNA(90%),Ki-67(20%) and p53(85%) expressions. After treatment with TNFα the positive percentage of expressions of PCNA,Ki-67,p21ras gene protain of CNE3 cells were decreased significantly(P<0.01) . There were no p53 positive cells, p21WAF1 /CIP1 positive cells were present instead in the CNE3 cells. Conclusion:TNFα could inhibit the growth of CNE3 cells by affecting the gene expression related to cell cycle progressing.
