摘要
目的 研究未分化状态的神经干细胞对机械损伤脊髓神经元的保护作用。方法 建立体外大鼠脊髓神经元机械损伤模型 ,在实验组中加入神经干细胞 ,对照组中加入无神经干细胞的培养液。用MTT比色试验、彗星试验、电镜检测神经干细胞对神经元的保护作用。结果 MTT比色试验表明实验组神经元活力明显高于对照组 ,彗星试验表明实验组DNA损伤程度较对照组轻 ,电镜显示实验组神经元细胞核、线粒体损伤减轻。结论 神经干细胞能有效保护机械损伤的脊髓神经元。
ObjectiveTo explore the protective effect of neural stem cells on mechanically injured spinal cord neurons.MethodsA traumatic insult was delivered to rat spinal cord neurons in culture by tearing the cell layer with a stylet in a grip pattern. Neural stem cells obtained from 1 day Sprague Dawley rat brain were added into spinal cord neuron cell culture at various concentrations and at different intervals following the mechanical insult. After 24 hours combined culture, MTT assay, comet assay and electron microscopic examination were carried out to observe the vitality, DNA damage, and ultrastructural changes of spinal cord neurons. The procedures were repeated to the cell cultrue in the control group where single culture media was added instead of neural stem cells.ResultsSpinal cord neurons of the experimental group had higher survival rate and lower DNA damage level than those of the control group, the differences being significant at higher concentrations of neural stem cells added at an early stage. Electron microscopic examinations revealed milder chromosome condensation of the nuclei and minor mitochondral swelling in the experimental group.ConclusionNeural stem cells can protect mechanically injured spinal cord neurons. The earlier the protection is applied, the more obvious is the result.
