摘要 【目的】分析肝细胞癌NKG2D表达及对NK细胞细胞毒的影响。【方法】应用免疫荧光技术和流式细胞术 (FCM )分选 2 5例肝细胞癌 (HCC)、10例健康对照的外周血NK细胞 ,定量分析NKG2D蛋白表达。用MTT比色法检测抗NKG2DpAb加入前后NK细胞的细胞毒效应。【结果】HCC患者NK细胞的NKG2D蛋白表达量均低于正常对照 ,差异有显著性 (P <0 .0 1)。抗NKG2DpAb能显著抑制NK细胞对K5 6 2、HepG2细胞的细胞毒效应 ,使NK细胞对两种靶细胞的细胞毒效应分别下降了 6 .7和 4 .1倍。【结论】NK细胞NKG2D表达异常与肝细胞癌的发生发展可能相关。 To analyze the expression of NKG2D and its effect on cytotoxicity of natural killer (NK) cells in human hepatocellular carcinoma(HCC).NK cells in the peripheral blood of 25 patients with HCC and 10 healthy subjects were sorted by immunofluorescence technique and flow cytometry .method (FCM). Expression of NKG2D protein was quantitatively analyzed .The cytotoxicity of NK cells before and after addition of anti-NKG2D pAb was detected by monotetrazolium (MTT) colorimetry.The levels of NKG2D expression in NK cells of HCC patients were significantly lower than those in normal controls (P<0.01) .The anti-NKG2D pAb could obviously inhibit the cytotoxicity of NK cells towards K562 and HepG2 cells and decrease their cytotoxicity to both target cells for 6.7 and 4.1 times, respectively. [Conclusion] Abnormal expression of NKG2D protein in NK cells may be correlated with the occurrence and progression of HCC.
