Smad-7与TGF-β_1蛋白在胆囊癌组织中的表达及意义

摘要 【目的】探讨Smad 7和TGF β1蛋白在胆囊癌组织中的表达及其意义。【方法】采用免疫组化SP法检测 4 9例胆囊癌和 2 0例慢性胆囊炎组织中Smad 7和TGF β1蛋白的表达 ,并分析两者的表达与胆囊癌临床病理特征的关系。【结果】Smad 7在胆囊癌组织中的表达阳性率为 6 7.3% (33/ 4 9) ,显著高于慢性胆囊炎的15 .0 % (3/ 2 0 ) (P <0 .0 5 ) ;胆囊癌组织中TGF β1蛋白的表达阳性率为 38.8% (19/ 4 9) ,低于慢性胆囊炎的80 .0 % (16 / 2 0 ) (P <0 .0 5 )。Smad 7的表达与胆囊癌Nevin分期和组织学分化程度有关 (P <0 .0 5 ) ,TGFβ1蛋白的表达则随胆囊癌Nevin分期进展、组织学分化程度的降低而逐渐下降 (P <0 .0 5 ) ,Smad 7和TGF β1蛋白表达之间呈负相关 (P <0 .0 5 )。【结论】Smad 7蛋白高表达和TGF β1蛋白表达下降可能参与了胆囊癌的发生、发展过程。 ObjectiveTo evaluate the expression of Smad-7(an effector molecule in TGF-β 1 associated signal pathway) and transforming growth factor-β 1(TGF-β 1 ) proteins in tissues of gallbladder carcinoma(GC) and its clinical significance.The expressions of Smad-7and TGF-β 1 proteins in tissues from 49 cases of GC and 20 cases of chronic cholecystitis were detected by immunohistochemical SP method, and the relation of both expression to the clinicopathological features of GC was analyzed.The positive expression rate of Smad-7 in GC tissues was 67.3%(33/49), significantly higher than that in chronic cholecystitis (15.0%,3/20)(P<0.05); whereas the positive expression rate of TGF-β 1 in GC tissues was 38.8%(19/49), obviously lower than that in chronic cholecystitis (80.0%,16/20)(P<0.05).The expression of Smad-7 was positively correlated with Nevin staging and degree of histological differentiation in GC(P<0.05);while the positive expression of TGF-β 1 decreased gradually accompanying the progression of Nevin staging and poor degree of histological differentiation (P<0.05). Negative correlation existed between the expression of Smad-7 and TGF-β 1 protein (P<0.05).[Conclusion]The high expression of Smad-7 and the decreased expression of TGF-β 1 may take part in the carcinogenesis and progression of GC.
