抗人纤维结合蛋白单克隆抗体的研制及其在两位点酶联免疫吸附分析中的应用

摘要 报道抗人纤维结合蛋白单克隆抗体的研制及其在两位点酶联免疫吸附分析中的应用。3D和12G两细胞株分泌的单克隆抗体的滴度分别为1:1000和1:1600(细胞培养液)。与人血浆中其它蛋白质成分无交叉免疫反应。本文ELISA的灵敏度为0.22±0.01mg/L。回收率95~105%。批内变异系数3.6~6.2%;批间变异系数5.4~5.6%。50例正常人血浆纤维结合蛋白的浓度为335±59.5mg/L(范围210~434mg/L)。 Monoclonal antibody (McAb) to human fibronectin was produced and used in two-site ELISA for measurement of human plasma fibronectin Two new McAbs to human fibtonectin named 3D and 12G were obtained. Titres of the two McAbs in cell culture media were 1:1000 and 1:1600 individuaUy.Western Blot test showed that there was no any cross reaction between 3D, 12G McAbs and other human plasma proteins except fibronectin The two-site ELISA was reliable with sensitvity of 0.22±0.01mg/L, recovery of 95~105%inte-rassay C.V.of 3.6~6.2% and intra-assay C.V. of 5.4~6.6%, The concentration of plasma fibronectin of 50 normal volunteers was 335±59.5mg/L(range210~434mg/L).
