摘要 目的 研究大蒜素对恶性疟原虫体外生长活性的影响。方法 用新鲜配制或储存的疟疾完全培养基大蒜素稀释液在不同浓度 (10 0 μg/mL~ 5 μg/mL)处理恶性疟原虫 ,实验过程中在 12h ,2 4h换液或者不换液。分别作用 12h ,2 4h和 48h后 ,检测虫体感染率的变化分析药物的作用效果。同时以大蒜素处理正常红细胞观察对虫体宿主细胞的影响。结果 以新鲜配制的大蒜素处理疟原虫时 ,作用 12h后 5 0 μg/mL的浓度即可清除虫体 ,当作用 48h后在低至10 μg/mL的浓度也可杀灭原虫。而使用储存大蒜素换液或者新鲜配制的大蒜素不换液则需要较高的浓度。大蒜素处理正常红细胞时 ,在高于 5 0 μg/mL的浓度对宿主红细胞有破坏作用 ,而该浓度下虫体生长可被有效抑制。 结论 大蒜素在体外具有强烈的抗疟作用。 3 0 μg/mL是可以有效抑制疟原虫生长但对宿主红细胞没有损伤的适宜浓度。 Objective To investigate the effects of allicin on the growth activity of Plasmodium falciparum in vitro. Method The falciparum malaria was treated with freshly prepared or stored allicin solution which was diluted by complete malaria culture medium, at different concentrations ranging from 100μg/mL to 5μg/mL, changing the medium at 12h and 24h or not, respectively. After the incubation of the parasite culture with the drug for 12h, 24h and 48h, respectively, the parasitemia was determined as an index of the drug effects. Meanwhile normal erythrocytes were treated with allicin to detect the effects on the parasite host. Result When treated with freshly prepared allicin solution the parasite was eliminated at 50μg/mL after incubation of 12h and at the concentration as low as 10μg/mL after 48h. And to reach the same effect higher drug concentrarion was in need when treated with stored allicin with changing medium or treated with freshly prepared allicin without changing medium. Treatment of normal erythrocytes led to the finding of disrupted erythrocytes at the concentration higher than 50μg/mL, and at this concentration the parasite growth was inhibited effectively. Conclusion Allicin has the strong antimalaria activity in vitro. 30μg/mL is an appropriate concentration that can effectively inhibit the parasite growth without damage to normal erythrocytes.
