摘要 目的 :探讨 β 淀粉样蛋白 (β amyloid ,Aβ2 5~ 3 5毒性作用能否诱导大鼠AD模型 ,以及热休克蛋白 70(heatshockproteins ,HSP70 )在海马神经元损伤中的表达。方法 :将 4 8只大鼠 ,随机分为 3组 (每组 16只 ) :AD模型组、溶媒体组、假手术组 ,前两组在双侧海马分别注射 2 μ1(10 μg2 )Aβ2 5~ 3 5、2 μl生理盐水 ,假手术组不注射任何溶液 ;于手术后 1、7、14及 2 1d采用免疫组织化学、积分光密度分析等手段对各组大鼠脑HSP70 的表达进行研究。结果 :AD模型组手术 7d后大鼠记忆明显减退 (P <0 .0 5 ) ,且HSP70 在海马神经元内表达ld时最高 ,7d锐减 ,14d和2 1d则逐渐减少。结论 :Aβ2 5~ 3 5可诱导大鼠AD模型 ,并对热休克蛋白 70在海马神经元损伤中的表达产生显著影响 ,HSP70 的表达是阿尔茨海默病 (Alzheimer’sdisease ,AD)大鼠脑内神经元存活的重要标志。 Objective: To investigate whether the toxicity of β-amyloid protein (Aβ_ 25~35) induces model of Alzheimer's disease and heat shock proteins expression 70 in adult ratbrain. Method: we used immunohistochemical and integral optical density analytic method to analyze the adult rat brains, which had been injected with Aβ_ 25~35 into the hippocampis. Result: expression of HSP_ 70 was detected at ld-7d-14d and 21d after operation in Aβ_ 25~35 injected ratbrains,but the level of HSP_ 70 drop off. Conclusion: Aβ_ 25~35 causes model of Alzheimer'sdisease and heat shock proteins 70 expression in adult rat brain neurons, expression of heat shockproteins 70 in adult rat brain neurons may be related to Alzheimer's disease pathological changes.
