摘要
从HL_(60)人早幼粒细胞白血病细胞株培养上清液(HL_(60)-SUP)中,经10%~50%饱和硫酸铵沉淀、DEAE-52(DE-52)阴离子交换柱层析、凝胶过滤及聚丙烯酰胺凝胶电泳(PAGE)分离出一种肿瘤免疫抑制因子(TDSF),分子量约为87kD(1D=0.992 1μ)。该因子是瘤细胞恶性增殖过程中的基因产物,但其分泌不完全被抗急非淋白血病药物所阻断。肿瘤细胞培养上清液无细胞毒作用。TDSF有兔疫抑制作用:能够抑制T淋巴细胞对PHA的增殖反应,抑制T淋巴细胞分泌IL_2,抑制外源性IL_2的淋巴细胞的增殖反应,其化学本质为蛋白质。
Tumor-derived immunosuppressive factor(TDSF) from culturesupernatant of HL60-human promyelocytic leukemia cell line was purified by precipitation with 10%-50% ammonium sulfate,DEAE-52 anionexchange chroma-tography, gel filtration and was identified by polyacrylamide gel electrophoresis. The results show that the molecular weight of TDSF is about 87kD. TDSF is a genetic product of tumor cells. The synthesis and secretion of TDSF from tumor cells were partially suppressed by drugs against acute non-lymphocytic leukemia. The culture supernatant of tumor cells was not cytoxic. TDSF was able to inhibit PHA-stimulated proliferation of lymphocytes, the IL-2 production of PHA-stimulated lymphocytes and the IL-2-dependent proliferation of activated lymphocytes. TDSF is protein in chemical nature.
