早孕绒毛Y基因的快速诊断

摘要 许多伴性遗传疾病需在孕早期进行胎儿性别鉴定,报告一种不经Southern转移的分子杂交方法,即利用凝胶原位杂交法对早孕绒毛Y基因进行快速诊断。该方法是在提取孕8周绒毛DNA,经微型琼脂糖凝胶电泳后,直接将凝胶抽干成凝胶膜,尔后以Y染色体3.4 kb DNA片段为特异性探针,直接在凝胶膜上进行分子杂交。本文成功地在48 h内对 1例DMD携带者早孕绒毛的胎儿性别进行了基因诊断。 The fetal sex should be detected in the early gestation for many inherited sex-linked diseases. One method of molecular hybridization, hybridized directly on agarose minigel without Southern transfer, was reported here to diagnose Y-gene from early gestational villi. By use of this method, DNA was extracted from early gestational villi (8 w), the agarose gel was pumped into a gel-membrane after electrophoresis, and then the sample DNA on gel membrane was hybridized direclty with specific PY3.4 probe. A fetal sex was successfully diagnosed by detecting DNA of early chorionic villi from the DMD carrier in 48 h.
