摘要
用健康人外周血淋巴细胞与雷公藤甲素(简称甲素)混合体外培养24 h,然后在光镜下和扫描电镜下观察甲素对淋巴细胞形态的影响。结果 0.5 mg%和 1 mg%甲素组淋巴细胞数分别为4.275×10~5±0.33/ml和2.950×10~5±0.19/ml;显微分光光度计测两组的淋巴细胞 DNA含量,其OD值分别为200.57±68.74和181.99±81.38,较对照组显著降低。培养后的淋巴细胞形态破坏。推测甲素通过干扰淋巴细胞DNA 合成抑制其免疫功能和损害其细胞结构。
Lymphocytes in peripheral blood of healthy humans were compounded and cultured 24 h with triptolide, and then morphological effects of trip-tolide on the lymphocytes were observed under a light and scanning electron microscope. The number of cultured lymphocytes of group 0.5 mg% and 1 mg% triptolide was 4.275 × 105 ± 0 .33/ml and 2.950 × 105±0 .19/ml respectively. The DNA level of cultured lymphocytes of the two groups was measured with a micro-spectrophotometer, and theirOD level was 200.57±68 .74 and 181.99±81.38 respectively. There was a significant decrease in the number and DNA contents of cultured lymphocytes as compared with the control group. Morphology of the cultured lymphocytes was destroyed. From the foregoiny it may be postulated that triptolide suppresses the immune function of lymphocytes and injures their cellular structure by interfering with the synthesis of DNA.
