穿心莲提取物抗血小板聚集作用的临床及实验研究 被引量:13

摘要 通过对31例健康志愿者和24例病人的研究,发现穿心莲(APN)提取物能明显抑制 ADP和肾上腺素诱导的血小板聚集反应,而对花生四烯酸和瑞斯托霉素诱导的血小板聚集反应无明显影响。APN对 ADP诱导的血小板5-HT 的释放也有明显抑制作用(P<0.01)。电镜观察表明,APN体外、体内均可抑制ADP 诱导的血小板致密颗粒和α颗粒的释放及管道系统的扩张。其作用机理可能是刺激血小板腺苷环化酶,提高血小板内cAMP水平,因而抑制了血小板聚集。 A significant inhibitory effect of andrographis paniculate ness (APN) extracts on platelet aggregation induced by ADP and adrenaline was observed in 31 healthy volunteers and 24 patients. The aggregation reactions induced by arachidonic acid (AA) and ris toncetin were not significantly affected. APN also exerted a marked inhibitory effect on the release of ADP-stimulated platelet 5-HT (P<0. 01). Electron microscope observation showed that APN could reduce the release reaction of dense and a-granules as well as the dilation of canalicular system both in vivo and in vitro. The mechanism of APN may lie in that it stimulates platelet adenylate cyclase and raises platelet cAMP level, thereby inhibiting platelet aggregation.
