血清铁蛋白放射免疫分析试剂盒的研制

摘要 按Frankel方法从人肝中提纯铁蛋白,亚基分子量20.5kD;免疫家兔制得抗血清,滴度在1:5×10~4以上,亲和常数Ka>1.2×10~9 l/mol;氯胺T法制得^(125)I-铁蛋白,放化纯度大于95%,比活度为1.30×10~6 Bq/μg,过量抗体结合率大于80%;建立放免分析方法,其灵敏度为1.9±0.5 ng/ml,批内变异CV<4.2%,批间变异CV<8.5%,平均回收率为98±5%;测定303例正常人及136例病人血清铁蛋白含量,结果与临床符合。 Ferritin was purified from human liver by Frenkel method. Molecular weight of ferritin subunit was 20. 5 kD. Rabbit serum of anti-human liver ferritin was prepared with the titre of more than 1 : 50000 and affinity constant K, of more than 1.2×10~9 1/mol. ^(125)I-ferritin prepared by chloro-amino T has the radiochemical purity of more than 955,specific activity of 1.3×10~6 Bq/μg and binding rate of superquantity antibody of more than 80%. Analytical method of RIA was established, the sensitivity, was 1.9±0.5 ng/ml, the rate of recovery Was 98±5%, the coefficients of variability for intraassay and interassay were< 4.2% and <8.5% respectively. Serum ferritin concentration was measured in 303 healthy persons and 136 relative patients. The results agree with the clinic diagnosis.
