摘要
目的观察丹玄口康注射液对大鼠口腔黏膜下纤维化成纤维细胞凋亡的作用机理。方法采用免疫组化测定法检测各组动物颊黏膜中成纤维细胞Bcl 2阳性细胞指数 ,并观察局部病理组织学改变。结果治疗 8周后 ,正常组和模型组比较 ,两者差异无显著性 (P >0 .0 5 )。丹玄口康组和雷公藤组Bcl 2阳性细胞指数明显低于模型组和正常组 ,其差异具有显著性 (P <0 .0 1 ) ,同时 ,丹玄口康组与雷公藤组比较 ,其疗效更有优势 ,其差异也具有显著性 (P <0 .0 5 )。结论丹玄口康通过改善局部血液流变学和微循环状态 ,明显抑制凋亡基因———Bcl 2基因 ,使局部Bcl 2含量减少而促使已发生纤维化的大鼠口腔黏膜成纤维细胞凋亡 ,从而抑制胶原在局部的堆积 。
Objective To investigate the effects of Danxuan Koukang (DXKK) injection on Bcl 2 expression of fibroblasts among rats with oral submucous fibrosis (OSF).Methods The model of OSF was prepared among rats at first.Then, DXKK injection was given to the animals in treating group (TG).By the end of therapy lasted for 8 weeks, the number of Bcl 2 positively expressed cells was determined by immunochemistry in the tissues of OSF and the positive indexes of this indicator were compared among TG,modeling group (MG),healthy group (HG) and positive controlling group (CG), supplemented with histopathological observation on these samples. Results There was no significant difference found between HG and MG in the positive index of Bcl 2 expression,while significant differences could be found among these two groups and treated groups with DXXK and positive controlling group respectively in this indicator.However, the effect of DXKK on the lesion was significantly superior to that of positive controlling one (P<0.05).Conclusions DXKK holds significant inhibiting effects on the expression of Bcl 2 in the tissues of OSF to induce apoptotic effects on fibroblasts.Then,OSF lesion should pe improved by reducing collegen depositing here.This may be brought about by improving micro circulation in diseased tissues. 〔
