IFN-α1b对人子宫内膜腺癌细胞系JEC增殖的影响及机制

摘要 目的 研究 IFN-α1b对子宫内膜腺癌细胞系 JEC增殖及细胞周期的影响。方法 不同浓度的IFN-α1b作用24h后,通过细胞计数观察对 JEC的抗增殖效应,应用流式细胞技术分析细胞周期改变和细胞周期调控因子 p21,cyclin A和cyclin E的表达。结果 不同浓度的 IFN-α1b对 JEC均有抗增殖作用,其中500 U/ml抗增殖作用最强。流式细胞仪(FCM)检测显示S期细胞积累,p21的表达增加和 cy clin A表达减少,而cyclin E的表达增加。结论 IFN-α1b能够抑制子宫内膜腺癌细胞系JEC的增殖,其机理可能与 p21的表达增加和cyclin A表达减少,导致S期细胞积累有关。 Objective To study the inhibitory effect of IFN-α1b on Endometrial adenocarcinoma cell line JEC and its mechanism by measuring the change of cell cycle and detecting the expression of p21, cyclin A and cyclin E.Methods After treated by IFN-α1b at different concentrations 24 hours, the amount of cell was measured by cell count to learn the effect of IFN-α1b on the proliferation of JEC cell; cell cycle phase and expression of p21, cyclin A and cyclin E were analyzed by flow cytometry(FCM). Results IFN-α1b all have inhibitory effects at different concertrations,the strongest of which is 500U/ml. FCM shows that cells are accumulated in S phase in cell cycle;the expression of p21 and cyclin E increase but cyclin A reduces.Conclusion IFN-α1b can inhibit proliferation of the JEC cell and its mechanism relates to cell arrest in S phase which is probably caused by the increased expression of p21 and the reduced expression of cyclin A.
