神经生长因子对新生大鼠缺氧缺血脑皮质保护作用的实验研究 被引量:1

The experimental research of NGF protective effect in hypoxic-ischemic brain cortex of neonatal rat

摘要 目的探讨缺氧缺血脑损伤对新生大鼠缺氧缺血脑损伤(HIBD)脑皮质NO含量,fasL蛋白表达的影响,及神经生长因子(NGF)对新生大鼠HIBD的干预治疗作用以及有关分子机制。方法建立HIBD动物模型后分NO含量测定组,fasL免疫组化测定组。每组又分为神经生长因子干预组、生理盐水对照组,假手术组,根据检测需要,在缺氧缺血后24h、3d、7d作脑皮质匀浆NO含量测定,fasL蛋白免疫组化SP法测定。结果假手术组、生理盐水组、NGF干预组脑皮质匀浆NO含量(pmol/L)测定依次为24h(4128±356)、(15963±432)、(12315±429);3d(4201±362)、(30122±459)、(5305±426);7d(4082±342)、(5126±428)、(4636±349)。FASL蛋白表达阳性率24h(805±358)、(1222±611)、(2108±512);3d(821±311)、(877±590)、(1350±422);7d(801±280)、(833±289)、(827±284)。结论新生大鼠HIBD后,结扎侧脑皮质NO含量增高,fasL蛋白的表达增高,神经生长因子能减少NO的产生量,抑制fasL蛋白的表达,对缺氧缺血脑皮质有保护作用。 Objective To discuss the effect of hypoxic-ischemic damage on NO content of neonatal rat brain cortex,fas-L protein expression,the interruption of NGF and its relative molecule theory.Methods The content of NO measurement group and fas-L immunohistochemical measurement group were divided after hypoxic-ischemic model was built.Each group was divided into NGF interruption group,normal saline control group and pseudo-operation group.According to the measurement purpose,to detect NO content and fas-L protein expression of brain cortex slurry in 24h,3d,7d after hypoxic-ischemic.Results The NO content of brain cortex slurry of pseudo-operation group,normal saline control group and NGF interruption group was as follows:24h:412.8±35.6,1596.3±43.2,1231.5±42.9;3d:420.1±36.2,3012.2±45.9,530.5±42.6;7d:408.2±34.2,512.6±42.8,463.6±34.9.The fas-L protein expression of three groups was as follows:24h:8.05±3.58,122.2±6.11,21.08±5.12;3d:8.21±3.11,87.7±5.90,13.50±4.22;7d:8.01±2.80,8.33±2.89,8.27±2.84.Conclusion After HIBD,the NO content and fas-L protein expression of ligation side of brain cortex rise.NGF can reduce the NO output,control fas-L protein expression and protect the hypoxic-ischemic brain cortex.
