摘要
采用HCl将蛋白质彻底水解的方法对苏云金芽孢杆菌HD - 1伴孢晶体的水解规律和酸水解条件下芳香族氨基酸的被破坏程度进行了研究 ,不经衍生HPLC法直接测定水解样中酪氨酸的含量。得出酸水解的最佳时间为 2 4h、水解剂的用量为 1.0mlHCl mg伴孢晶体、水解最佳温度为 110℃。
Parasporal crystal of Bacillus thuringiensis HD-1 was hydrolyzed completely with HCl,tyrosine in the hydrolysate was determined directly by HPLC.The hydrolytical laws of Bt HD-1 and the destroyed degree of aromatic amino acids when they were hydrolyzed were studied.The following conclusions on circumfluence conditions were drawn:hydrolysis time is 24h,volume of HCl is 1.0ml per mg parasporal,and hydrolysis temperature is 110℃.The study about hydrolysis's law of parasporal crystal will help to the determination of toxin of Bt.
