摘要
以酿酒酵母SaccharomycescerevisiaeHY0 1为出发菌株 ,分别对其进行化学诱变和紫外诱变。选用 5 -溴尿嘧啶和盐酸平阳霉素两种低毒性诱变剂联合使用 ,通过均匀设计确定了两种诱变剂的最佳配比即 5 -Bu浓度为 30 0 μmol L、平阳霉素浓度为5 μmol L时 ,获得一定数量的正突变株 ,最高海藻糖含量是 15 .98%。在紫外诱变过程中经摇瓶初筛、复筛获得了高产海藻糖菌株 ,其海藻糖含量为 19.5 2 % ,比出发菌株提高了 35 %。
Saccharomyces cerevisiae HY01 was used as the starting strain,and was treated with chemical revulsant and UV irradiation.Two revulsants which contain low toxicity were taken into the experiment together,and by uniformity design we got the best quota of them,that is 5-Bu 300μmol/L and Pingyangmycin hydrochloride 5μmol/L.After chemical mutagenesis we got some plus mutants of which the highest yielding is 15.98%.After first and second screen during UV mutagenesis process we obtained a high trehalose producing strain which produces 19.52% increasing 35% as compared with the starting strain.
