前列腺癌p53、bcl-2、bax基因表达与凋亡、增殖的关系 被引量:3

摘要 目的 研究前列腺癌组织中细胞增殖与细胞凋亡 ,及其相关蛋白表达意义。方法 采用原位细胞凋亡标记技术(TUNEL)及免疫组织化学ABC法对 3 6例前列腺癌 (Pca)和 11例正常前列腺 (NP)组织石蜡切片 p5 3、bcl 2、bax、PCNA蛋白及细胞凋亡检测。结果 前列腺癌细胞的增殖指数和细胞凋亡指数较NP明显增高 ,且AI/PI比值较正常组织AI/PI低 (P <0 0 1) ;随着肿瘤分级的增加 ,细胞增殖水平明显增加 (P <0 0 1)。p5 3蛋白表达与前列腺癌细胞增殖水平相关 ;bcl 2蛋白表达与细胞凋亡水平相关 (P <0 0 5 ) ;bax基因表达与凋亡无关 ,但发现它们的bcl 2 /bax的比值与凋亡有关 ,结果显示 :高bcl 2 /bax比值多见于低凋亡组 ;低bcl 2 /bax比值多见于高凋亡组。结论 细胞增殖与细胞凋亡均参与了前列腺癌的发生、发展。p5 3基因与细胞增殖水平的调控有关。bcl 2和bax在细胞凋亡调节中起到重要作用 ,由于bcl 2蛋白过量表达引起bcl 2 /bax失平衡 ,在前列腺癌发生、发展过程中起到重要作用。 Objective To investigate relationship between proliferatio n apoptosis and expression of related genes in prostate cancer. Methods The expression of p53, bcl-2, bax and PCNA protein and apoptosis cell were detected by immunohistochemical method and in site end labeling method (TUN EL) respectively in tissues of 36 cases of prostate carcinoma and 11 cases of no rmal prostate (NP). Results The result showed that there was significant difference in both proliferative and apoptotic indexes between the t umor and normal tissues(P<0.05), but the rate of AI/PI of the prostate car cinoma was higher than that of normal prostate (P<0.01). The increment of proliferative index was positively correlated with pathological grade(P<0. 01). The expression of p53 and bcl-2 was separately correlated well with prolif erative index and apoptosis index. Conclusion The results sug gest that proliferation and apoptosis was both involved in the development of th e prostate cancer. p53 was correlated with the modulation of proliferation respe ctively; bcl-2 and bax play an important role in apoptosis regulation, the imba lance of bcl-2/bax mainly induced by the undue expression of bcl-2 plays an im portant role in development of prostate cancer.
