DHA高产菌Schizochytriumsp.FJU-512脂肪酸组成与18S rRNA基因序列分析 被引量:6

摘要 采用松花粉垂钓法分离到一株docosahexaenoicacid (DHA)高产菌FJU - 5 12。该菌株DHA含量高 (占总脂肪酸的 5 6 .2 4 % ) ,其它长链杂酸含量少 (仅有Docosapentraenoicacid ,二十二碳五烯酸 ,DPA) ,极具开发应用价值。高密度培养获得 33g L生物量。该菌株呈二分裂生长 ,没有分生孢子。对其 18SrRNA基因进行了克隆测序并登录GenBank(AY75 8384 )。依据 18SrRNA基因建立的系统进化树表明 A strain FJU-512 producing high concentration of docosahexaenoic acid (DHA) was isolated by using pine pollen as bait. The oil within single cell was very valuable for containing polyunsaturated fatty acid up to 56.24% of the total fatty acids with only less docosapentaenoic acid(DPA) but DHA. The cells growth was dependent on binary division without conidia. The 18S rRNA gene was successfully isolated by PCR and sequenced (GenBank: AY758384).The phylogenetic tree based on 18S rRNA gene showed that Schizochytrium sp.FJU-512 was close to Schizochytrium limacinum.
