蝎毒抗肿瘤多肽对小鼠肝癌细胞H_(22)及移植瘤的抑制作用 被引量:3

摘要 目的观察纯化蝎毒抗肿瘤多肽穴APBMV雪,<3500kD灌胃对小鼠肝癌H22动物移植瘤的抑制作用及对H22细胞的体外抑制杀伤作用。方法将接种H22的带瘤小鼠分为对照组、环磷酰胺治疗组和纯化蝎毒抗癌多肽治疗组。其中APBMV浓度1.2mg/ml、0.8mg/ml,以0.5ml灌胃,给药10d后处死,取实体瘤称重并计算肿瘤生长抑制率。采用噻唑蓝(MTT法)、集落形成抑制实验,用丝裂霉素C(MMC)为阳性对照药品,以蝎毒提取物SVCⅠ、SVCⅡ、SVCⅢ、SVCⅣ各组分(浓度分别为45μg/ml、60μg/ml、75μg/ml、90μg/ml),分别处理人大肠癌细胞H22,观察H22的生长情况。结果APBMV在应用的前两个剂量范围内对H22移植瘤有明显的抑制作用,抑制率大于50%,与对照组比较差异显著(P<0.05),三个不同剂量组之间差异亦显著,有明显的量效关系。SVCⅢ对Lovo细胞的细胞毒性和集落形成的抑制作用略强于MMC组,与对照组差异显著穴r=0.98熏P<0.01雪,SVCⅠ、SVCⅡ、SVCⅣ对细胞H22的影响,在所用药物浓度内与对照组比较,无显著性差异,细胞毒性作用不显著。结论口服APBMV能有效的抑制H22移植瘤的生长和对H22细胞具有明显的细胞毒性和生长抑制作用。 Objective To observe the inhibitory effect of antineoplastic polypeptide from Buthus martensii venom (APBMV) on transplanted tumors-hepatoma 22(H22) and its cells. Methods Tumor weight, tumor growth inhibitory rate(IR%) of tumor bearing mice were used as the indexes. Mice were inoculated by H22, and 24h late the tumor bearing mice were divided into control, CTX-treated and the APBMV-treated groups which were administered 1.2mg/ml,0.8mg/ml,0.4mg/ml,0.5ml of APBMV separately by po. CTX-treated and control group were given 0.2mg/g of CTX and equal volune saline,respectively.Tumor bearing mice were killed after administrating the chemicals for 10 days.And using the MTT colorimetric method and the colony formation test as the indexes, the mitomycin C(MMC) as the positive control chemical, Lovo cell line was treated with SVCⅠ-Ⅳat different final concentration of 45靏/ml,60靏/ml,75靏/ml,90靏/ml. The effect of SVCⅠ-Ⅳ on Lovo cells were observed at different intervals after the cells were exposured to SVC.Result Growth of H22 was inhibited markedly by APBMV at three doses used in this test. Inhibitory rate was over 30%compared with control group and among three doses there was a significant difference (P<0.01), and the SVCⅢ had obvious cytotoxicity on the Lovo cell .There were obvious dose-response relationship in test of SVCⅢ(r=0.84,P<0.05). SVCⅢ also possessed inhibitory effect on colony formation of Lovo cells,with good dose-response relationship(r=0.98,P<0.01).Conclusion Growth of H22 tumors and the cell were efficiently inhibited by APBMV, which also has cytotoxic effect on the cell.
