摘要  目的 研究在不同放射性浓度下, 5 125I 2′ 脱氧尿嘧啶核苷(125I UdR)对大鼠C6 神经胶质瘤细胞的杀伤效应,并初步探讨其发生机制。方法 在大鼠C6神经胶质瘤细胞的培养液中,加入不同放射性浓度 125I UdR培养 24h后,通过细胞形态学变化、细胞存活实验及流式细胞仪检测细胞凋亡率进行观察。结果 在加入 74MBq/L放射性浓度的 125IUdR培养 24h后,细胞形态上表现出皱缩、变圆、脱落现象,贴壁细胞数目减少。不同放射性浓度的细胞存活分数 (SF)随着浓度升高明显下降,同一浓度下与对照组相比较,存在统计学意义。同时流式细胞仪 (FCM)检测随着 125I UdR的升高,细胞凋亡率增加, 24h细胞凋亡率最高接近 20%。结论 125I UdR对大鼠C6 神经胶质瘤细胞具有明显的杀伤效应,并随着浓度的提高而增强,其发生机制可能是通过诱导细胞凋亡而产生。 Objective To study the lethal effect of the various radioactive concentration of 125 I UdR on rat C 6 glioma cells.Methods Rat C 6 glioma cells were cultured with medium of the various radioactive concentrations for 24 hours, then the lethal effect was observed through surviving fraction and rate of apoptosis estimated by flow cytometry (FCM). Results After being cultured with 74 MBq/L radioactive concentration of 125 I UdR for 24 h, morphology of cells changed to circling, shrinking, abscission and the number of seeding reduced. Surviving fraction decreased with increasing of radioactive concentration. In comparison, there was statistically significant difference in cells treated with 125 I UdR. Meanwhile, rate of apoptosis through FCM increased with radioactive concentration. The top rate of apoptosis approached 20%. Conclusion The lethal effect of 125 I UdR on rat C 6 glioma cells is observed and it is increased with increases in radioactive concentration of 125 I UdR. The mechanism of cell killing may be due to apoptosis.
