摘要
目的 探讨酵母菌多聚糖(zymosan)对气道上皮细胞系A549作用以及气道上皮细胞参与呼吸道疾病的可能机制。方法 通过培养获得持续传代的A549细胞,并将A549细胞与相应刺激物共培养 24或 48h后,收获培养悬浮液,检测其中白细胞介素(IL 6)和IL 8水平。结果 A549细胞可以自发释放IL 6和IL 8;A549细胞接受酵母菌多聚糖刺激后释放的IL 6和IL 8显著性增多,并且随刺激物剂量增加而增加。刺激时间延长也可增加IL 6和IL 8的表达。在接受同样酵母菌多聚糖刺激后TNF α预刺激的A549细胞释放的IL 6(P<0 001)和IL 8较未预刺激的A549细胞显著性增多。结论 在接受酵母菌多聚糖刺激时,呼吸道上皮细胞可能通过激活IL 6和IL 8信号通路释放IL 6和IL 8,从而参与气道疾病的发生。
Objective The interaction of resident cells in the airways with inhaled allergens, may result in local airway inflammation in pre disposed atopic individuals. The epithelium of the airways plays a key position to participate in such a local event, which may thus contribute to the pathogenesis of airway disease. In this study, we focused on the effects of zymosan on A549 cells and the possible mechanism behind them. Methods A549 cells were kept serial passage by cultured in RPMI1640 with heat inactivated fetal calf serum at 37℃ with %5 CO 2. A549 cells were seeded in 24 well sterile plates and went on to be challenged with stimuli. After 24 h or 48 h co culture with stimuli the supernatant was harvested and kept in -20℃ for further measurement of IL 6 and IL 8. Results A549 cells released IL 6 and IL 8 spontaneously and released significantly increased IL 6 and IL 8 when challenged with zymosan. TNF α pre activated A549 cells had more potent to release IL 6 ( P <0 001)and IL 8 ( P =0 045 ) when challenged with zymosan compared to rest A549 cells. Conclusion Airway epithelial cell play an important role in resident local inflammation partly through signals of IL 6 and IL 8 in zymosan induced inflammation.
