摘要 目的 向人外周血来源的树突状细胞 (DCs) 转染人MUC1质粒, 使DCs特异性的表达和提呈MUC1。方法 应用脂质体将人MUC1质粒转染体外培养的外周血来源的DCs, RT-PCR检测转染人MUC1质粒DCs的表达。结果 转染MUC1质粒的DCs, RT-PCR检测有一特异性扩增带。结论 用脂质体能成功地将人MUC1质粒转入DCs并表达。 Objective To detect the specific expression and presentation of MUC1 in the human blood-derived dendritic cells(DCs) which transected with plasmid MUC1.To prepare the vaccine of DCs transected with plasmid MUC1.Methods MUC1 was successfully transected into cultured human blood-derived DCs with lipofectin.MUC1 mRNA expression in DCs〔pcDNA3 1(+)-MUC1〕 was detected by RT-PCR.Results MUC1 mRNA expression in DCs was confirmed with RT-PCR.Conclusion MUC1 in the DCs 〔pcDNA3 1(+)-MUC1〕 is successfully expressed with lipofectamine.
