脂肪酸类药物诱导分化治疗脑胶质瘤分子机制研究 被引量:3

Study on the relevant molecular mechanisms of differentiation-inducing therapy for brain glioma with fatty acid analogs

摘要 包括苯乙酸、苯丁酸和丙戊酸在内的脂肪酸类药物对多种肿瘤细胞具有一定的诱导分化作用。在胶质瘤方面,它们不但可以抑制细胞增殖和诱导细胞形态向良性方向改变,而且能使细胞内的过氧化物酶体增殖物激活受体(PPAR)、组蛋白去乙酰化酶(HDAC)、激活蛋白-1(AP-1)等基因表达量发生变化,从而调控细胞分化。本文对脂肪酸类药物的药理作用、疗效及作用机制进行介绍。 Analogs of fatty acids including phenylacetic acid, phenylbutyric acid and valproate has certain differentiation-inducing effects on various kinds of tumor cells. In glioma, they can not only inhibit cell proliferation and induce tumor cells toward benign differentiation, but also change the expression of peroxisome proliferator-activated receptor(PPAR), histone deacetylase (HDAC), activating protein-1(AP-1),which may be the key gene in controlling the differentiation-inducing effect of fatty acids on glioma in vivo. In this review, we introduce the pharmacology,the curative effect and the potential mechanism of fatty acid analogs.
