大鼠脑缺血再灌注损伤后死亡相关蛋白激酶的表达及意义

Expression and significance of DAPK after cerebral ischemia reperfusion in rats

摘要 目的 探讨大鼠局灶性脑缺血再灌注损伤时死亡相关蛋白激酶 (Deathassociatedrelatedproteinkinase ,DAPK)的表达变化及其与脑缺血再灌注损伤的关系。方法 线拴法建立局灶性脑缺血再灌注模型 ,SD大鼠随机分为正常对照组、假手术组和缺血再灌注组 ,缺血组于栓塞 3h后分别再灌注 12h、2 4h、3d、7d。HE染色观察组织病理变化 ;免疫组化法检测DAPK的表达 ;原位杂交法检测DAPKmRNA的表达。结果 缺血组DAPK免疫阳性反应于再灌注后 2 4h开始增强并于 3d时达高峰 ,随后下降 ;DAPKmRNA的表达于再灌注 3d时达高峰 ,随后下降。结论 脑缺血再灌注损伤诱导DAPK表达增强 ; Objective To explore the alterations of death-associated related protein kinase (DAPK) expression and its relationship to cerebral ischemia reperfusion injury in rats.Methods The focal cerebral ischemia/reperfusion models of rats were achieved by middle cerebral artery occlusion (MCAO).The adult rats were randomly divided into normal control,sham-operation and ischemia/reperfusion groups.The ischemic group was respectively reperfused for 12h,24h,3d and 7d after ischemia for 3h.Pathological changes of injured neurons were demonstrated by H-E st aining,the expression of DAPK was assayed by immunohistochemistry and the expression of DAPK mRNA was detected by in situ hybridization.Results The expression of DAPK was strongly increased at 24h and peaked at 3d after reperfusion,then decreased.The peak expression of DAPK mRNA occurred at 3d after reperfusion.Conclusion These observations support that the expression of DAPK increases significantly in the cerebral tissues after reperfusion of focal cerebral ischemia and strongly supports a key role for DAPK in ischemia/reperfusion brain injury.
