IL-1ra抑制兔后发性白内障的实验研究

目的 探讨IL 1ra(白细胞介素 1受体拮抗剂 )对兔眼晶状体囊外摘除术后IL 1等炎性细胞因子及后囊膜混浊的影响。方法 哈白兔 4 0只 ,随机分为 5组 :对照组 (单纯行晶状体囊外摘除术 ) ,结膜下给药组 ,前房内给药组分 3组 ,2 5 μg/ml、5 0 μg/ml、10 0 μg/ml。术前、术后 3天、7天、14天、2 8天取泪液 ,采用放射免疫法检测IL 1、IL 2、IL 6、TNF α含量。分别于术后 4周、8周应用裂隙灯照相观察后发性白内障的形成情况。结果 对照组兔泪液中IL 1、IL 2、IL 6、TNF α含量在术后 3天、7天、14天较术前增高。给药组术后 3天、7天、14天IL 1、IL 2、IL 6、TNF α的水平较术前不同程度增高 ,但峰值明显降低。各组泪液中 4种细胞因子的含量于术后 2 8天基本恢复到正常。给药组后囊膜混浊在术后 8周时与对照组有显著性差异。结论 兔眼晶状体囊外摘除术后IL

Objective To explore the effect of IL-1ra on inflammatory cytokine like the IL-1 etc and posterior capsular opacification(PCO) in eyes of rabbits.Methods Forty rabbits were randomly divided into five groups.Control Group(rabbit eyes were only given ECCE surgery),injected Subconjunctiva Group,injected into anterior chamber Group (including 25μg/ml,50μg/ml,100μg/ml three sub groups).Collected tears at pre-operation and 3 days,7 days,14 days,28 days after operation,using RIA method to examine IL-1,IL-2,IL-6,TNF-α content.Use slip-lamp digital photo investigated the condition of PCO afteroperator for 4 and 8 weeks,respectively.Results The content of IL-1,IL-2,IL-6,TNF-α after operation 3 days,7 days,14 days was higher than those of pre-operation in rabbit's tears.The conditions of giving IL-1ra groups' s density after operation was higher than those of pre-operation too,but clearly lower than those of control groups's density after operation 3 days,7 days,14 days in rabbit's tears.Every group's content of the four kind of cell factor was renew normal after operation 28 days.The PCO was different between control group and giving IL-1ra groups after operation 8 weeks.Conclusion IL-1 and other cell factors have important effect on forming PCO as inflammation medium after ECCE operation in rabbits's eye.
