摘要
来自拟南芥的rd2 9A基因受干旱、高盐及低温诱导表达 ,编码一种与LEA蛋白相似的亲水性很强的蛋白。用Rd2 9A启动子控制GUS报告基因的表达构建载体 ,通过基因枪介导转化小麦幼胚愈伤组织 ,经过分子生物学检测及在PEG胁迫条件下的GUS组织化学染色证实了rd2 9A启动子在小麦愈伤组织中能够诱导GUS基因的表达。
rd29A gene from Arabidopsis, encoding LEA-like protein, was induced by stresses like drought, high-salt and low-temperature. An expression vector containing GUS gene under control of rd29A promoter was constructed and bombarded into wheat immature embryogenic calli by biolistic. The results of PCR, PCR-Southern and histochemical stain showed that expression vector was transferred into wheat calli and GUS gene was induced by PEG treatment.
