摘要
目的探讨大鼠颈动脉球囊导管扩张术后,损伤动脉内皮细胞的再生动力学规律。方法建立大鼠颈动脉气囊导管损伤模型,伊文思蓝染色标记缺乏内皮覆盖的血管区域,动态观测内皮细胞再生规律;免疫组织化学染色观测损伤后不同时期动脉壁内TGFβ1的表达情况;计算机图像分析技术检测损伤动脉内皮细胞再生部位与乏内皮覆盖区内膜及中膜增生面积。结果术后9d前内皮再生缓慢,术后9 ̄28d再生速度较前显著增高(1.19±0.14)mm/dvs(0.44±0.25)mm/d,P<0.01,术后28d后内皮再生趋于停滞。术后早期(9d以内)TGFβ1在动脉内壁内的高表达,而14d后,TGFβ1表达显著减少。结论PTA术后早期内皮再生受抑制,早期再生速度较慢,9d后再生速度明显加快,28d后再生趋于停滞。术后早期(9d以内)TGFβ1在动脉壁内的高表达可能是内皮再生受抑制的直接原因之一。
To investigate the kinetics of endothelial cells (Ecs) regeneration after balloon angioplasty in rats. The de-endothelium region of injured artery was indicated with Evan's blue staining, and the endothelial cells regeneration at different postoperative time was measured. The expression of TGFβ1 in local vascular tissue was detected with immunohistochemistry. We found that the Ecs regeneration was obviously slow in the first 9 d and became remarkably faster during postoperative 9d to 28 d in contrast (1.19±0.14) mm/d vs (0.44±0.25) mm/d,P <0.01. Meanwhile, the expression of TGFβ1 had a negative correlation with the pattern of ECs regeneration in vivo. [Conclusion] Our result suggests that ECs regeneration after balloon injury is inhibited at early period. The high expression of TGFβ1 potentially contribute to inhibit the ECs regenration.
