摘要
目的:通过研究异常瘢痕患者外周血单个核细胞HLA-DR,-DQ,-DP及CD1a分子的表达及分布,探讨免疫遗传因素在异常瘢痕形成中的作用。方法:采用免疫细胞化学方法检测10例瘢痕疙瘩患者、10例增生性瘢痕患者、10例扁平瘢痕患者和10例正常人外周血单个核细胞HLA-II类分子(HLA-DR,-DQ,-DP)及CD1a分子的表达及分布。结果:①HLA-DR+,-DQ+,-DP+及CD1a+细胞数量在瘢痕疙瘩患者、增生性瘢痕患者、扁平瘢痕患者和正常人各组间差异无显著性意义(P>0.05)。②瘢痕疙瘩和增生性瘢痕组外周血HLA-DR+细胞(813.16±62.77,883.11±146.71;F=10.613,P<0.01,-DQ+细胞(420.12±94.25,439.24±146.65;F=5.143,P<0.01),-DP+细胞(259.40±57.43,271.93±70.38;F=9.177,P<0.01)及CD1a+细胞(580.15±108.48,648.94±185.46;F=8.581,P<0.01)的积分光密度高于扁平瘢痕和正常人组外周血HLA-DR+(636.22±133.95,597.88±166.36),-DQ+(302.77±89.77,308.57±44.05),-DP+(192.96±51.32,159.01±31.99)及CD1a+(423.53±142.36,368.67±117.18)细胞的积分光密度,表明瘢痕疙瘩患者和增生性瘢痕患者组外周血单个核细胞HLA-DR,-DQ,-DP及CD1a蛋白含量高于扁平瘢痕患者和正常人组。结论:瘢痕疙瘩患者和增生性瘢痕患者外周血HLA-DR,-DQ,-DP及CD1a蛋?
AIM:To investigate the role of immunogenetic factors in the pathogenesis of abnormal scars based on the research of the expression levels and distribution patterns of HLA Ⅱ (HLA DR, DQ, DP) and CD1a molecules in peripheral blood mononuclear cells of patients with abnormal scar.METHODS:The expression levels and distribution patterns of HLA Ⅱ (HLA DR, DQ, DP) and CD1a molecules in peripheral blood mononuclear cells were determined in 10 samples of keloid,10 samples of hypertrophic scar,10 samples of thin and flat scar and 10 cases of normal people with immunocytohistochemical staining.RESULTS:① The amounts of HLA DR+ , DQ+ , DP+ and CD1a+ cells had no significant differences among the keloid,hypertrophic scar,thin and flat scar and normal people groups(P >0.05).② The integrated absorbance of HLA DR+ cell(813.16± 62.77,883.11± 146.71;F=10.613,P< 0.01), DQ+ cell(420.12± 94.25,439.24± 146.65;F=5.143,P< 0.01), DP+ cell(259.40± 57.43,271.93± 70.38;F=9.177,P< 0.01) and CD1a+ cell(580.15± 108.48,648.94± 185.46;F=8.581,P< 0.01) molecules in the keloid and hypertrophic scar groups was higher respectively than that of HLA DR+ (636.22± 133.95,597.88± 166.36), DQ(302.77± 89.77,308.57± 44.05), DP+ (192.96± 51.32,159.01± 31.99) and CD1a+ (423.53± 142.36,368.67± 117.18) molecules in the thin and flat scar and normal people groups.This indicates that the contents of HLA DR, DQ, DP and CD1a protein in the keloid and hypertrophic scar groups were was higher than those of thin and flat scar and normal people groups.CONCLUSION:The increased contents of HLA DR, DQ, DP and CD1a protein in peripheral blood of patients with the keloid and hypertrophic scar suggest that the hyperplasia of scars may be associated with immunogenetic factors.
