凋亡抑制基因Survivin在宫颈癌中的表达及其生物学特性

摘要 目的:研究表明抗凋亡基因Survivin在恶性肿瘤中表达增高,故探讨Survivin基因在宫颈癌中的表达与生物学特性的关系,为预测宫颈癌的预后提供实验依据。方法:研究于2001-03/2003-07在哈尔滨医科大学分校附属医院妇产科和第三附属医院妇科完成。应用RT-PCR的方法检测51例宫颈癌组织及10例正常宫颈组织Survivin的表达。结果:Survivin蛋白表达阳性率随宫颈癌的病理分级及临床分期的增高而表达增强。淋巴结转移组阳性率为86.2%,高于无淋巴结转移组的阳性率(45.4%),二者差异有显著性意义(P<005)。结论:Survivin基因在宫颈癌中高表达,与癌组织分化程度、临床分期、淋巴结转移呈正相关,与病理类型无关。可作为判断宫颈癌发生、发展、转移等生物学行为的一个指标。 AIM:Researches indicate that the expression of anti apoptotic gene Survivin in malignant tumor is increased. This paper investigates the expression of Survivin gene in cervical cancer and its relationship with biological characteristics, so as to provide experimental evidence for predicting the prognosis of cervical cancer. METHODS:The study was done in the Department of Obstetrics and Gynecology,Affiliated Hospital, Branch School of Harbin Medical University,and the Department of Gynecology,Third Affiliated Hospital of Harbin Medical University from March 2001 to July 2003.The expression of Survivin gene was detected in 52 cervical cancer tissues and 10 normal cervical tissues by using reverse transcription polymerase chain reaction(RT PCR). RESULTS:The positive rate of Survivin protein expression was enhanced with the increases of pathological grade and clinical stage of cervical cancer.The positive rate of Survivin expression in the lymph node metastasis group(86.2% ) was higher than that in the group without lymph node metastasis(45.4% )(P< 0.05). CONCLUSION:The high expression of Survivin gene in cervical cancer is positively correlated with the degree of cancer histological differentiation,clinical stage and lymph node metastasis,but has no relation with the histological type.It can be used as one of the indexes for judging the biological characteristics of occurrence, development and transfer of cervical cancer.
