摘要
目的 建立一种简便、快速的胶体金免疫渗滤法 (DIGFA)用于鼠疫菌的快速诊断。方法 用柠檬酸钠还原法制备 10nm的胶体金标记鼠疫多价抗体 ,制备成鼠疫抗体金探针。结果 鼠疫胶体金渗滤法对鼠疫FI抗原的最低检出限为 10ng/ml;对EV76的最低检出限为 2 .4× 10 5cfu ;除 4株鼠疫菌外 ,不与包括小肠结肠炎和假结核耶尔森菌等其他细菌发生交叉免疫反应 ;整个反应过程在 10min内完成。结论 该方法简便、快速、特异、敏感 ,不需任何特殊仪器设备 ,适合于现场鼠疫监测。
Objective A rapid,simple dot immunogold fltration assay(DIGFA) was established to detecting Yersinia pestis.Methods 10 nm colloidal gold was made by deoxidizing method with sodium citrate,anti-Plague polycolnal antibody was marked with 10 nm colloidal gold to prepare solloidal probe.Results The DIGFA could detect concentration of antigen as low as 10 ng/ml and its sensitive was 2.4×10 5 cfu;the whole detecting time was less 10 min;except 4 Y strains of the four biotype,the DIGFA had no cross immune reaction with other 7 strains including 1 Y enterocolitica strain and 1 Y pseudotuberculosis strain.Conclusions The DIGFA is a simple,rapid,specific,sensitive and reliable assay for detecting Y pestis on spot without special apparatus.
