摘要
目的 探讨碘缺乏 (低碘 )、甲状腺功能减退 (甲减 )大鼠仔鼠海马组织c -fos、c -jun的表达。方法 分别选用低碘饲料及他巴唑诱导建立低碘及甲减大鼠仔鼠动物模型 ,取生后 2 0、3 0d仔鼠海马免疫组织化学S -P法染色 ,对c -fos、c -jun表达产物进行图像分析。结果 生后 2 0d低碘、甲减组仔鼠海马组织c -fos灰度值分别为 162 .76± 1.0 8、15 9.80± 3 .85 ,显著高于对照组 15 0 .91± 4.95 (P <0 .0 1) ;c -jun灰度值分别为 14 6.87± 1.97、14 0 .2 7± 12 .11,也显著高于对照组 13 0 .71± 8.73 (P <0 .0 1)。生后 3 0d低碘、甲减组仔鼠海马组织c -fos灰度值分别为 162 .98± 0 .83、163 .2 9±1.5 2 ,显著高于对照组 15 3 .3 2± 4.70 (P <0 .0 1) ;c -jun灰度值分别为 14 5 .80± 1.3 9、14 6.5 7± 1.5 1,明显高于对照组12 5 .2 9± 3 .5 1(P <0 .0 1)。结论 碘缺乏、甲状腺功能减退可降低仔鼠海马组织原癌基因c -fos、c -jun表达 ,进而影响神经系统及智力发育。
Objective To study the expression of c-fos and c-jun in developing hippocampus of iodine deficiency and hypothyroid rat offsprings.Methods The models of deficient iodine and hypothyroid rats were established by pre-treatment with low iodine diet and Thiamazole respectively.The expression of c-fos and c-jun in hippocampus tissue on postnatal day 20,30 were studied by means of S-P immunohistochemical technique and micrograph analysis system.Results The means of average gray value of c-fos in the iodine deficiency and hypothyroid groups on postnatal day 20(162.76±1.08,159.80±3.85) were significantly higher than that in the control group(150.91±4.95)(P<0.01),the means of average gray value of c-jun in the iodine deficiency and hypothyroid groups(146.87±1.97,140.27±12.11) were significantly higer than that in the control group(130.71±8.73)(P<0.01).The means of average gray value of c-fos in the iodine deficiency and hypothyroid groups on postnatal day 30(162.98±0.83,163.29±1.52)were significantly higher than that in the control group(153.32±4.70)(P<0.01),the means of average gray value of c-jun in the iodine deficiency and hypothyroid groups(145.80±1.39,146.57±1.51) were significantly higher than that in the control group(125.29±3.51)(P<0.01).Conclusion Iodine deficiency and hypothyroidism of mother rat may impair the expression of both c-fos and c-jun of their offsprings.
