反相高效液相色谱法测定大血藤中大黄素的含量 被引量:3

Quantitative determination of emodin in the vine of Sargentodoxa cuneata by RP-HPLC

摘要 目的建立反相高效液相色谱法测定大血藤中大黄素含量的方法.方法选用DiamonsilTMC18色谱柱(200 mm×4.6 mm,5 μm);流动相:甲醇-0.1 %磷酸水溶液(80:20);检测波长:254 nm;流速:1.00 mL/min;柱温:35 ℃.结果大黄素进样量在0.0416~0.2080 μg范围内线性关系良好,r=0.9997,平均加样回收率为101.4 %,10批大血藤饮片中大黄素的含量范围为6.72~13.71 μg/g.结论该方法操作简便,快速,准确,重现性好. Objective To establish a RP-HPLC method for quantitative determination of emodin in the vine of the plant of Sargentodoxa cuneata (Oliv.) Rehd. et Wils. Method DiamonsilTM C 18 column (200 mm×4.6 mm, 5 μm) was used as the chromatographic column, and methanol-0.1% aqueous solution of phosphoric acid was used as the mobile phase; the detection wavelength was 254 nm; the flow velocity was 1.00 mL/min; and the temperature of the column was 35℃. Result Emodin showed a good linear relationship when the amounts of the sample were between 0 0416~0 2080 μg (r=0.9997), the average recovery was 101.4%, and the range of the content of emodin was found to be 6.72~13.71 μg/g.Conclusion This RP-HPLC method is simple, fast and accurate, with good repeatability.
