不同年龄人鼻中隔软骨细胞生物学特性观察 被引量:2

摘要 目的观察年龄因素对体外培养的人鼻中隔软骨细胞生物学特性的影响。方法体外培养不同年龄组人鼻中隔软骨细胞,观察其生物学形态、细胞增殖周期、甲苯胺蓝异染和Ⅱ型胶原的免疫组化染色,并测定35S-Na2SO4掺入量以观察对软骨细胞蛋白多糖合成量的影响,在细胞特性和增殖特征方面进行比较。结果各组间软骨细胞的形态、传代特征及传代数、组织学检测无明显差异,细胞倍增时间随年龄增大而下降,软骨细胞蛋白多糖合成量随体外时间延长而上升,不依赖年龄。结论人鼻中隔软骨细胞生物学特性总体上不依赖年龄,其作为软骨组织工程的潜在种子细胞来源具有广泛的年龄段。 OBJECTIVE To study the effect of do- nor age on biological character of human nasoseptal chondrocytes cultured in vitro. METHODS The com- parison was done in cellular and proliferation charac- teristics of cultured chondrocytes isolated from 24 pa- tients ranging in age from 15 to 60 years. This study observes the morphology of the chondrocytes and shows the histologic characteristics by toluidine blue and type Ⅱ collagen immunohistochemistry reaction, and determines proliferation of chondrocytes by dou- bling time. The proteoglycan synthesis efficiencies of donor age on human nasoseptal chondrocytes cultured in vitr o were m easured by using 35 S-Na2SO4 incorporation. RESULTS The results suggest that hu- man nasoseptal chondrocytes isolated from different age still show the similar morphology and histologic character, while doubling time decrease with age ncreasing, and proteoglycan synthesis increase with time in vitro but show independence with age. CON- CLUSION Many cellular characteristics seem to be independent of donor age for the span from 15 to 60 years. The use of human nasoseptal chondrocytes as a source of cells for tissue engineering may be valid over a wide range of patient ages.
