脂质体转染人IFN-β基因诱导胶质瘤细胞SHG44凋亡的实验研究

摘要  目的 探讨人β干扰素(IFN β)与胶质瘤细胞凋亡的关系及意义。方法 利用脂质体转染方法将IFN β真核表达载体pSV2IFN β导入SHG44胶质瘤细胞。细胞免疫荧光和免疫组化检测IFN β基因的稳定转染及表达,利用流式细胞仪、透射电镜观察细胞凋亡情况。结果 IFN β基因成功转染SHG44胶质瘤细胞并得以表达。转染细胞的凋亡细胞数与未转染细胞相比有显著性差异。结论 IFN β能够诱导人SHG44胶质瘤细胞凋亡。 Objective To elucidate the relationship between interferon β(IFN β) and glioma cell apoptosis.Methods Eukaryonic expression vector (pSV2 IFN β) for IFN β was transduced into glioma cell SHG44 by using lipofectamine.The stable transfection and expression of IFN β in the glioma cell SHG44 were determined by immunohistochemistry and fitc.Cell apoptosis was also observed by flow cytometry and electronic microscopy.Results IFN β gene was successfully transfected and expressed in glioma cell SHG44.There was significant difference in the number of apoptosis cells between transfected cells and non transfected cells.Conclusions IFN β can induce glioma cell SHG44 apoptosis.
