卵母细胞发育潜能与颗粒细胞凋亡关系的初步研究 被引量:6

摘要 目的探讨卵母细胞发育潜能与颗粒细胞凋亡之间的关系.方法将卵母细胞在体外培养28~30 h后,取其颗粒细胞,采用TUNEL法进行凋亡率的分析,并与卵子的成熟情况和卵子的评分及形成胚胎结果进行对照. 结果经培养后,随着卵母细胞成熟度的降低,凋亡率升高;而且随着卵母细胞评分和发育潜能的降低,颗粒细胞凋亡率逐渐升高.结论卵子发育潜能与颗粒细胞凋亡密切相关,通过测定颗粒细胞的凋亡率可预测未成熟卵母细胞的发育潜能. Objective: To investigate the relationship between the developmental ability of human oocytes and the apoptosis of granulosa cells. Methods: After in vitro maturation for 28~30 hours, all the granulosa cells of each human oocyte were collected and the rates of apoptosis in granulosa cells were analyzed by terminal deoxynucleotidyl transferase mediated dUTp nick end labeling (TUNEL). The scoring of immature oocytes before culture, the maturity of the cultured oocytes and subsequent fertilization and cleavage of the oocytes were observed. Results: With the decreased maturity of corresponding oocyte, the rate of apoptosis in granulosa cells was increased. As the scoring and developmental capacity of the oocyte was decreased, the rate of apoptosis in granulosa cells was increased. Conclusion: The relationship between the developmental capacity of the oocyte and apoptosis of granulosa cells was significant. The apoptosis rate of granulose cells could be a predictor of the developmental potential of the corresponding oocytes.
