摘要
目的研究新型光敏剂CDHS801在膀胱癌T24细胞内的亚细胞定位。方法将传代培养的膀胱癌T24细胞与CDHS801共同孵育,采用激光共聚焦显微镜为成像工具,应用线粒体荧光探针MitoTrackerREDCMXRose和MitoTrackerGreenFM来进行CDHS801的亚细胞定位分析。结果激光共聚焦显微镜下CDHS801、MitoTrackerREDCMXRose和MitoTrackerGreenFM发出的荧光都位于细胞浆内,在细胞核周围呈点状散在分布。结论CDHS801经膀胱癌T24细胞摄取后,定位于细胞浆内,主要分布线粒体内。
Objective To discuss the accurate subcellular localization of CDHS801 in bladder carcinoma T24 by using laser scanning confocal fluorescence microimaging system.Methods Laser scanning confocal fluorescence microimaging system was applied and T24 cell was incubated with CDHS801.MitoTracker RED CMXRose and MitoTracker Green FM were selected to label mitochondria.Fluorescence of CDHS801,MitoTracker RED CMXRose and MitoTracker Green FM were collected through corresponding emission filter.Results The fluorescence of CDHS801,MitoTracker RED CMXRose and MitoTracker Green FM was all detected in the cytoplasm of the T24 cell,outside the nucleus indicating the diffuse intracellular distribution of the photosensitizer and the mitochondria probe.Conclusion CDHS801 distributes in the cytoplasm,mainly in the mitochondria,little in the nucleus.
