啤酒酿造用凝集性酵母融合育种的研究 被引量:8

摘要 用红霉素抗性突变和小菌落呼吸缺陷突变,标记一株非凝集性啤酒酿造用酵母 Sacch(?)-romyces carlsbergensis B8然后与凝集性酵母 Saccharomyces carlsbergensis A43进行电诱导融合,获得五株稳定的凝集性酵母融合株。其中,F6、F7具有与亲株 A43同等强的凝集特性,同时还保留了另一亲株 B8的发酵力强,酿造风味好等生产性状。经测定细胞的 DNA含量,五株融合株分别属于二、三、四倍体。用 Octyl-sepharose CL-4B 疏水柱层析法测定细胞表面的疏水性表明,A43和全部融合株的细胞表面疏水性能都比非凝集性亲株 BB强,并与细胞的凝集性能呈正相关。 Five flocculent brewery yeast fusants were obtained by protoplast fusion method.The fu-sants were obtained from a strain of Saccharomyces carlsbergensis A43 and a respiratory de-ficient petites of S.carlsbergensis B8 with erythromycin-resistant property.The later is aBurns Value Test negative non-flocculent brewing yeast.The fusants F6,F7 acquired the floc-culent capacity as its parent A43 an retaining the industrial fermantative merit of another parentstrain B8.The cell DNA content determination of the fusants showed their ploidies are 2n,3n and4n respectively.The cell surface hydrophobicity has been determined by Octyl-sepharose CL-4Bhydrophobic interaction chromatography.The results showed A43 and all of the fusants aswell displayed stronger hydrophobic property than their parent strain B8.And the cell hydro-phobicity is in accordance with their flocculence intensity.
