丹参提高胸水肿瘤浸润淋巴细胞杀瘤作用的研究及其超微结构的改变

STUDY ON EFFECTS OF TUMOR INFILTRATING LYMPHOCYTE IN PLEURAL FLUIDON TUMOR CELL ENHANCED BY RED SAGE INJECTION ANDCHANGES OF ITS ULTRSTRACTURE

目的 :观察丹参注射液提高胸水肿瘤浸润淋巴细胞的杀瘤作用及其超微结构改变。方法 :从癌症患者恶性胸水中分离获取TIL和自体瘤细胞 ,体外培养观察丹参注射液对TIL体外长期存活 ,扩增及抗肿瘤作用的影响 ,并着重观察了其杀瘤作用的形态超微结构改变过程。结果 :丹参注射液可使TIL体外长期存活。扩增后的TIL在体内外的抗肿瘤活性均明显增强 ,作用类似IL - 2。光电镜观察到扩增后的TIL与自体瘤细胞共同培养具有使癌细胞裂解作用。结论 :功能和形态二方面证实丹参注射液具有明确的抗肿瘤免疫作用 ,是较为理想的替代细胞因子、辅助TIL细胞过程免疫治疗恶性肿瘤的免疫调节剂。

Objective:To observe the enhancement effect of red sage injection on tumor infiltrating lymphocyte (TIL) in pleural fluid to kill tumor cell and the changes of its ultrastructure.Methods:TIL and the self-tumor cells were separated and obtained from the malignant pleural fluid of cancer patients,secondly cultivated in vitro to observe the effects of red sage injection on the long-term growth,amplification and anti-tumor function of TIL,emphatically observe the changes of its ultrastructure. Results:Red sage injection could make TIL survive for a long time in vitro,the anti-tumor activity of amplified TILS enhanced obviously both in vitro and vivo,its function was similar to IL-2.The amplified TILS which were cultivated with self-tumor cells could make tumor cell split and collapse by light microscope and eletron microscope.Conclusion:Red sage injection has definite anti-tumor immune action,which has been manifested from function and appearance. It is much better the replacement to cytokine and the immune regulator of assisting to cure malignant tumors with the immunization therapy of TIL.
