氯氮平对雄性小鼠血糖和胰岛素的影响

Effects of clozapine on blood glucose and serum insulin in male C57BL/6 mice

目的 :探讨氯氮平对雄性C5 7BL/ 6小鼠血糖和胰岛素的影响。 方法 :6 3只雄性C5 7BL/ 6小鼠随机分为 3大组 ,空白组、氯氮平 4mg/kg组、氯氮平 2 0mg/kg组 ,于灌药后的 3h、1周、4周测定空腹血糖、糖耐量、胰岛素。 结果 :灌药后 3h、1周空腹血糖、血胰岛素、血糖曲线下面积都无显著升高 ;灌药 4周后空腹血糖值显著升高。腹腔注射高糖后 6 0min的氯氮平 2 0mg/kg组的血糖值及血胰岛素显著升高。 结论 :氯氮平可以慢性升高小鼠的空腹血糖和胰岛素 ,影响糖耐量 ,但急性期无明显影响。

Objective:To investigate the effects of clozapine on blood glucose and serum insulin in the male C57BL/6 mice. Method:63 male C57BL/6 mice were randomly divided into three groups,which were administrated with distilled water, clozapine 4 mg/kg, clozapine 20 mg/kg. The mice were tested the fasting blood glucose,glucose tolerance and serum insulin after the treatment of three hours,one week,four weeks respectively. Results:The fasting blood glucose,serum insulin and glucose AUC had no significant increase after the administration of three hours and one week.However,after four weeks,the fasting glucose increased significantly.After the treatment with glucose (25% 2 g/kg ip) for 60 minutes, the blood glucose and the serum insulin in clozapine 20 mg/kg group increased significantly. Conclusion:It is demonstrated that clozapine increase serum insulin and the fasting blood glucose slowly and influence glucose tolerance,but the immediate influence on blood glucose and serum insulin might be little.
