摘要
选用未经产雌性金黄地鼠30只,分别于第8,8.5,9,10,11天颈椎脱臼处死,剖腹取胎,固定于10%福尔马林,常规石蜡包埋,连续切片,应用免疫组织化学 ABC 法,观察和分析心血管酸性成纤维细胞生长因子(aFGF)的分布状况。结果显示:aFGF 广泛分布于心内膜、心肌和心内膜垫间充质,第8.5天阳性着色最强,随着胚龄增加,免疫染色逐渐减弱。光镜下可见,aFGF 分布于心内膜垫间充质细胞、内皮细胞的细胞质和细胞外间质中,细胞核呈弱阳性着色。实验表明,aFGF 在胚胎早期心血管发育过程中发挥着重要作用,并与细胞生长、分化有关。
30 virgin females of the gold hamster were killed at 8,8.5,9,10,11day,respectively,preg- nant uteri with embryos were fixed by 10% formalin,routinely embeded with paraffin and sec- tioned in series.Sections were stained with immunohistochemical method(ABC)for observation of distribution of aFGF in the embryonic heart.The results show that immunoreactive aFGF was i- dentified widely in endocardium,myocardium and the mesenchymal cells of cardiac cushions,and Immunoreactive of aFGF changes with increasing cell diferentiation.At the cellular level,aFGF was localised in the cytoplasm as well as extracellular matrix,and faint staining was also noted in some nuclei.The above results suggest that aFGF play an important role in the development of mouse heart,and is associated with cell growth and differentiation.
