摘要 运用正交实验设计探讨黄连CoptischinensisFranch.中小檗碱的提取工艺。由极差分析和方差分析得知,在本实验考察的影响因素中,温度对提取的影响最大,固液比对提取的影响次之,提取次数对提取的影响最小。实验结果确定黄连中小檗碱的最佳提取条件为:温度为60℃,9倍体积的50%乙醇,提取2次,每次1h。在此条件下,黄连中小檗碱的提取率达到91%以上。分光光度法测定小檗碱的平均回收率为97 38%。6次平行的相对标准偏差CV为1 38%。 The optimal extraction process for berberine in rhizoma of the Coptis chinensis Franch. was investigated by orthogonal method. By the variance analysis, the temperature , extraction times and ratio of solid-liquid had different effects on extraction. Among three factors, temperature had an obvious effect on extraction. The results showed that on the condition of temperature 60℃, 1∶9 of 50% ethanol to the rhizoma of Coptis chinensis Franch. extraction for 2 times and 1h each time,the average extraction rate was as high as 91%. The average recovery rate of berberine determined by spectrophotometry reached 97.38% and the coefficient of variation was 1.38%.
