摘要 [目的]观察全蝎提取液对急性早幼粒细胞株(HL-60)细胞及相关基因的影响。[方法]全蝎诱导HL-60细胞凋亡的形态学观察及流式细胞仪分析;全蝎对HL-60细胞周期分布的影响;免疫组化法观察全蝎对HL-60细胞凋亡调控基因表达的影响。[结果]全蝎可以明显促进HL-60细胞的程序化死亡,并在一定范围内呈正比关系,最高浓度(10mg/mL)时,凋亡细胞比例明显增加,达21.44%。中、小剂量药物浓度时,细胞凋亡比例略有增加,分别为12.56%和9.47%,对照组仅7.36%。同时可显著提高P53基因的表达水平(与对照组相比,P<0.01)和降低bcl-2基因的表达。[结论]全蝎提取液对HL-60细胞凋亡及相关基因的表达具有益影响。 Objective]To observe the effect of Scorpio extract on progranulocyte(HL 60) in acute promyelocytic leukemia and the correlative gene. [Methods]Scorpio induced morphologic changes of apoptosis of HL 60 and flow cytometry analysis, the effect on the distribution in cellular cycle of HL 60, the effect on the expression of modulating genes in apoptosis of HL 60 determined with immuohistochemistry were investigated. [Results] Scorpio could markedly accelerate the programming death of HL 60. The positive dose effect correlation could be found in certain concentration range. At the highest concentration(10 mg/mL) the proportion of apoptosis increased obviously, peaking at 21.44%. When middle or small dosage were administered, the proportion increased slightly. They were 12.56% and 9.47% respectively. It was only 7.36% in control. Simultaneously, it could markedly increase the expression of P53 gene(P<0.01 compared with control),and decrease the expression of bel 2. [Conclusion]Scorpio extract has beneficial effect on HL 60 death and correlative gene expression.
