摘要
目的:研究1,6 二磷酸果糖(FDP)对大肠杆菌内毒素(ET)致大耳白兔急性肺损伤(ALI)的保护性 作用及其对磷脂酶A2(PLA2)活性的影响。方法:大耳白兔随机分为对照组、ET致伤组、ET致伤+FDP干预组。 经静脉一次性注射ET复制兔ALI模型。各组分别于0h、0.5h、2h、4和6h等不同时点测定呼吸频率、心率、血 压、血气分析及血清中PLA2活性;于6h点处死动物,测定肺组织PLA2活性,并行肺组织病理学观察。结果:静注 ET后,致伤组动物与对照组相比较,出现呼吸频率加快、血压下降、心率加快、血氧分压降低和PLA2活性增高,肺组 织出现明显的病理损害。干预组动物肺损伤轻于致伤组。结论:FDP对PLA2激活起抑制作用,可改善其病理生理 过程;对ET所致的兔早期ALI有一定的保护作用。
Objective:To study the effects of Fructose-1,6-diphosphate(FDP) on Endotoxin(ET)-induced acute lung injury(ALI) and the activities of phospholipase A 2(PLA 2) in rabbits. Methods:Japanese flap-eared white rabbits were randomly assigned to three groups: control group (group A), ET-treated group (group B) and combination group(treated by ET and FDP, group C).Acute lung injury was induced by the injection of ET at a time. During the experiment, respiratory rates, heart rates, blood pressure, arterial blood gases and the plasma PLA 2 activities were measured at 0h, 0.5h, 2h, 4h and 6h respectively. Then, after rabbits being killed at 6h, the pulmonary PLA 2 activities were measured, and the pathologic changes were examined with light microscope. Results:Compared with group A, rabbits of group B showed the typical characters of ALI, including respiratory rates and heart rates increasing, as well as blood pressure and arterial blood oxygen pressure decreasing. Significant increase of the PLA 2 activities was also seen both in blood serum and lung tissue in group B. Conditions in group C were better than those in group B. Conclusion:FDP can inhibit the PLA 2 activities, improves its pathologic physiological procedure, and possesses protective effects on early ALI in rabbits induced with ET.
