水杨酸钠对肾性高血压大鼠耳蜗Hsf1 mRNA表达的影响 被引量:1

摘要 目的观察水杨酸钠对肾性高血压大鼠耳蜗Hsf1表达的影响 ,并分析其可能的机制。方法将造模 4周后模型成功大鼠 36只 ,另设正常对照组 (假手术组 ) 12只 ,分为 4组 ,每组 12只 ,A组 :正常对照组 (假手术组 ,Sham组 ) ;B组 :模型对照组 (2K1C组 ) ;C组 :2K1C +水杨酸钠组 ;D组 :2K1C +水杨酸钠 (45 0mg·kg-1·d 1) +L 精氨酸(L Arg ,2 0 0mg·kg-1·d 1)组。采用RT PCR方法检测耳蜗组织Hsf1mRNA表达情况 ,并做半定量法分析。结果① 2K1C +水杨酸钠 +L Arg尾SBP较模型对照组和 2K1C +水杨酸钠组明显降低 (P <0 .0 5 ) ;②正常对照组Hsf1mRNA有微弱表达 ,模型对照组Hsf1mRNA表达较高 ,2K1C +水杨酸钠组表达最高 ,2K1C +水杨酸钠 +L Arg治疗组表达较弱 ;OD值比较分析发现 ,模型对照组Hsf1mRNA表达较正常对照组Hsf1mRNA表达有显著增高(P <0 .0 1) ,2K1C +水杨酸钠组与模型对照组比较有显著性差异 (P <0 .0 5 ) ,2K1C +水杨酸钠 +L Arg治疗组OD值明显低于模型对照组 (P <0 .0 5 )以及 2K1C +水杨酸钠组 (P <0 .0 1) ,2K1C +水杨酸钠 +L Arg治疗组OD值与正常对照组比较无显著性差异 (P >0 .0 5 )。结论水杨酸钠可提高耳蜗组织Hsf1mRNA表达 ,L Arg可使水杨酸钠所致的耳蜗组织Hsf1mRNA表达下调。 Objective To study the effects of sodium salicylate on the expression of Hsf1mRNA in cochlear tissues of rats with renovascular hypertension and explore the possible mechanism.Methodes Thirty-six healthy rats were divided into three groups(B:2K1C group;C:2K1C+ sodium salicylate group ;D: L-Arg group) and another 12 rats were added as control group (group A).The animals were sacrificed. After L-Arg was given for four weeks.The expression of Hsf1 mRNA was detected by semi-quantitative RT-PCR in cochlear tissues of rats with renovascular hypertension.The animals were sacrificed after salicylate was given for four weeks.The mRNA expression of Hsf1mRNA was detected by semi-quantitative RT-PCR in cochlear tissues of rats with renovascular hypertension.Results ①SBP of D group was markedly decreased than group B and C.②RT-PCR analysis showed expression of Hsf1mRNA in cochlear tissues of rats with renovascular hypertension.The expression of Hsf1mRNA in group A was weak experssion;while those of group B and C were high;the expression of Hsf1mRNA in group D was remarkablely decreased than those of group B(P<0.05) and C (P<0.01).Conclusion Sodium salicylate could enhance the expression of Hsf1mRNA in cochlear tissues of rats with renovascular hypertension,while the expression of Hsf1mRNA could be decreased by L-Arg.
