家兔微卫星标记群体遗传变异快速分析方法的建立 被引量:12

摘要 目的分析VcⅠ系獭兔(VcⅠ)、VcⅡ系獭兔(VcⅡ)、青紫蓝兔(QZL)、新西兰兔(NZR)和日本大耳白兔(JWR)5个品种(系)家兔群体内遗传变异和群体间遗传距离。方法每个品种(系)兔随机抽取16只,综合利用微卫星多重PCR、DNA池、复合点样和快速银染技术。结果建立起了一套快速分析家兔群体遗传变异的方法。5个品种(系)家兔群体内遗传变异情况为VcⅡ>VcⅠ>JWR>QZR>NZR,聚类结果为VcⅠ与VcⅡ亲缘关系最近,与JWR、QZL亲缘关系较近,与NZR亲缘关系最远。结论实验所得的各群体家兔遗传变异结果与Vc獭兔实际育种事实相符。 Objective To analyse the population genetic variation in 5 breeds of rabbits as Vc Ⅰ, Vc Ⅱ, QZL, NZR, JWR, and the genetic distance among them. Method 16 rabbits were selected from each population, and the technology of multi PCR,DNA pool,multiple loading,speedy silver staining was used. Results\ A set of speedy method of analyzing the population genetic variation of domestic rabbits was constructed. The order of genetic variation value was that: Vc Ⅱ>Vc Ⅰ>JWR>QZL>NZR. The result of cluster displayed that the population whose relationship was nearest to Vc Ⅰ Rex rabbit population was Vc Ⅱ Rex rabbit population, followed by Japanese white rabbit population, QZL rabbit population. New Zealand rabbit population was the furthest one to Vc I Rex rabbit population in relationship. Conclusion\ The result of genetic analysis in this study was conformed to the actual fact of the breeding history of the Vc Rex rabbit.
